DNA Mismatch Repair Deficiency in Colorectal Adenocarcinoma and its Association with Clinicopathological Features

چکیده
Background: Evaluation of mismatch repair (MMR) status in colorectal cancer (CRC) could be used as the guidance for postsurgical treatment. Defective MMR status is associated with distinct clinicopathological features. This study aims to evaluate associations between MMR status and clinicopathological features in a retrospective cohort of consecutive Chinese CRC patients. Methods: Clinicopathological information was collected from 481 consecutive individuals who underwent curative surgery for CRC. MMR status was determined by Immunohistochemistry (IHC) staining, which was used to examine expression of MLH1, PMS2, MSH2 and MSH6 on paraffin embedded tissues containing adenocarcinoma. Results: IHC results showed that 33 (of 481, 6.9%) CRC cases presented loss of MMR proteins expression, including 26 (of 205, 12.7%) from colon and 5 (of 262, 1.9%) from rectum. Tumors with deficient DNA mismatch repair (dMMR) status were significantly related to younger patients, larger tumor size, mucinous-focal or signet-ring histological phenotype, and preferred right-sided location (P<0.001). And dMMR tumors also implied poor differentiation and more occurrence in stage CRC patients. No significant association was found between MMR status and gender, invasion depth and lymph node metastasis. Conclusions: We concluded that the prevalence of abnormal MMR protein expression in this cohort of Chinese CRC patients was comparable with the West. Meanwhile, tumors with dMMR status were associated with distinct clinicopathological characteristics of CRC patients.

Mutational analysis of the BRAF gene in colorectal mucinous carcinoma in association with histological configuration.
Genetic alterations and their association with clinicopathological features in colorectal mucinous carcinoma (MC) remain unknown. In particular, little is known about the mutational status of the BRAF gene, which is activated by oncogenic Ras. This study aimed to evaluate the status of BRAF together with K-ras, p53 and mismatch-repair deficiency to clarify their association with tumorigenesis o...

Mismatch repair (MMR) genes play an important role in the occurrence and development of sporadic colorectal cancer; however, the effect of MMR genes on clinicopathological features and prognosis remains unclear. The aim of the present study was to observe the clinical significance of MMR gene expression in sporadic colorectal cancer. Clinicopathological data and postoperative samples from 404 p...
متن کاملExpression of cytokeratin 20 indicates invasive histological phenotype in poorly differentiated colorectal adenocarcinoma.
BACKGROUND/AIM Cytokeratin (CK) 20 expression is an independent prognostic factor of poorly differentiated adenocarcinoma (PDA) of the colon and rectum. We aimed to investigate the mechanism of its involvement through a clinicopathological study. PATIENTS AND METHODS We analyzed 156 surgically resected PDAs, which were sub-classified as solid type (Por1) showing expansive growth, or non-solid...
متن کاملClinicopathological Characteristics and Prognosis of Colorectal Cancer in Chinese Adolescent Patients.
BACKGROUND Colorectal adenocarcinoma rarely occurred in adolescent. Clinical feature and prognosis of this population are not clear until now. In addition, DNA mismatch repair (MMR) status may relate to the early disease occurrence. The present study aimed to perform a retrospective analysis of adolescent patients with colorectal cancer, including clinicopathological characteristics and prognos...
متن کاملSerrated adenocarcinoma morphology in colorectal mucinous adenocarcinoma is associated with improved patient survival
Colorectal mucinous adenocarcinoma (MAC) and serrated adenocarcinoma (SAC) share many characteristics, including right-side colon location, frequent mucin production, and various molecular features. This study examined the frequency of SAC morphology in MACs. We assessed the correlation of SAC morphology with clinicopathological parameters, molecular characteristics, and patient prognosis.
